29G Biocube filter mod. Will it cycle?

Currently I have a 29G Biocube with 20 lbs of live rock and 20 lbs of live sand. The tank has been up and running for about four months. (since Labor Day) Three fish and a few zoas/polyps for livestock.

In the second chamber (middle) I have two Eshopps sponges and a bag of Seachem Matrix Bio Media (1L). I just ordered the inTank Media Basket and a Tunzee 9001 protein skimmer.

I am wanting to remove the sponges and Matrix and add the skimmer and media basket (sponge w/ filter floss/Purigen/Chemi-Pure Elite) to the middle chamber.

My question: Will this cause my tank to cycle? I didn't think it would with all the live rock I have but I'm second guessing at the last minute.
