The Top Rim of my 29 gallon Biocube is leaking - HELP

The leak is small right now, but it is persistent. It appears to be in one small spot and not quite as wide as a small drinking straw. I got an Aquarium Safe Silicone but, do I need to drain the entire tank to seal the rim. I'm worried about leaving the fish and coral out of water while it cures. I'm really new at this, obviously, so I apologize if these questions seem stupid to you veterans.

Can I take the water to just below the water level, apply the sealant, then fill it back up? The silicone sets in 5 minutes but cures completely in 24 hours.

How long should I wait before putting water back in to the correct level?

Do I need to seal both sides (which if I do answers how long I need to wait) or just the outside where the plastic rim overlaps the glass? Will an airstone provide enough circulation while I let it cure overnight?
