I put the rock in my tank, followed by sand and then water to ensure that the rock had good contact with the glass. I want to move it around now though. Is that alright? I doubt i'll get good glass contact now that the sand is in there.

You don't have much choose, but if your sand bed is not too deep, maybe scoot the rock over, bulldozing the sand, or use a Turkey baster and blow the sand away around the rock and then move the rock, returning the sand afterwards.
