I’ve used hot glue for stuff had no I’ll effects
So maybe another member has information on it

Idk what that piece is, but I wouldn't glue it in. Looks like it will get dirty and clogged often. I wouldn't use it at all

Not sure what that is, but hot melt glue degrades and does not last long immersed in salt water.
It's okay if it's not getting wet.

Silicone is a sealant not an adhesive.

it’s a plug hole but every time I plug it in it keeps falling off so idk if I should put glue on it or silicone

If it's underwater I'd use some sort of screw bulkhead with a gasket on each side.
Depends on how much pressure it's under.

Low pressure you could use aquarium silicon sealant with the plug on the inside so the pressure is pushing it tighter to the glass.
