I found a piece of floating (what looks to be) a zoa polyp. When I took it out of the water, it was mushy and round. I couldn't tell which end is up, or if it's even a zoa polyp.

Decided to glue it to a plug anyway.

I am wondering if I did happen to glue it upside down, would it still be OK or did I just kill it?

Hard to say. when I find a single polyp blowing around I try to put it in a lighted crevice, ive also put them in plastic cups weighted with sand, like with mushrooms.

They take hold rather quickly. I think as long as there is something for it to grow out of it has a fighting chance.
