After I add water to your system for the first time, should I wait for the sand to settle before powering up the pumps and powerheads or start them up right away?

I turned mine on when I added water. If you are using a filter or anything similar you might want to keep that off so it doesn't pick up anything.

I'm going to be using a foam filter block in my sump

I would just turn it on. Leave skimmer off till it settles but that's just me. Someone else might chime in.

When I cycled my last tank with a hob filter I just turned on everything. Got a little noise from the pump on the filter but I just cleaned it out and it's still going.
