With just that one? Can you pull the rock out, as in, is it an "on top" rock, and not supporting other rocks, because if that's the case, I would put some tank water in a 5 gallon bucket, put the rock in it, and treat that spot with kalk paste.

Bummer. You can kalk paste it in the tank, I would just prefer to take it out so that you can kill it and make sure it doesn't spread, and your puffer doesn't investigate the new pasty area.
