I am putting together ~50 gallon FOWLER tank (52in x 31in x 12in). I have never done FOWLER before so some insight on stocking would be helpful.

So far my list consists of:
Royal gramma
2x clowns
Maybe a dwarf angel if they can do well long term in my size tank

I have rock seeding as well as about 1000g of filter media that has been seeding in my other tank for a good while. I have two left over canister filters (I know they’re not the best but from what I’ve read they can do well in FOWLER when maintained regularly?)

I’m not looking for a crazy stocked tank but I do want some variation in colour if I can get it.

Start with the Clowns, Royal Gramma are nice but a little aggressive if introduce first
Try and get fishes that work I like fox face, Dejardini are very good fish and keeps your rock clean

If you're sure you're not going to have coral, consider getting some fish that are not reef safe (or reef safe with caution). Blue spotted puffer, dwarf/fuzzy lionfish, or a flameback angelfish.

I am very certain this tank will not have coral in it, I have a mixed reef already and I’m not looking for another reef at the moment. Would a flameback angle become aggressive with the stocking I mentioned?

It's always hit or miss but with a flameback angel as long is it's the only angel in there it should be fine. Some people have had royal grammas that are a menace, but mine is a complete wimp and will bolt into the rockwork the second I start walking towards the tank. Personalities can always change, so be prepared to have to catch a fish and either put it into a different tank or an acclimation box. I recently had to remove most of my rockwork to catch one of my clownfish because she was relentlessly bullying the other clownfish.

I'm a fan of FOWLR tanks, because I like fish more than Corals.
I like the color variations on some of the fish that you can put in there
My 50 gallon FOWLR has this stocking list. Flame Angel, Mocha Clown, Pajama Carinal, Six Line Wrasse, Yellow Tail Damsel, Coral Beauty Angel, Klein Butterfly, Royal Gama and Valentini Butterfly.
They all get along and there's a lot of movement and color with this group.
