So my skimmer is L10.25 x W5.5 x H20.75

I'm designing a 40b with its own skimmer section and the current dimension for that section is L10.5 x W6.5 H8

Will this be enough room for the skimmer to work properly?

As long as neither the input or output is blocked (and the skimmer physically fits), it will be fine....although you aren't leaving much wiggle room.

What I will did to my diy sump is that I put my skimmer in the tank 1st then I took the measurement then I set it up.

The skimmer picture is before I moved the baffles.

Thanks for the info

It's going in a baffled section in a 40B.
I'm copying/modifying a racetrack design sump that has 4 sections
- drain/filter sock - left section
- fuge/frag grow out - front section
- skimmer - rear right corner/section
- return section - rear middle section

I think I may try to rework the design to give the skimmer a 1" buffer on all sides, it should help reduce build up of grime and make it easier to clean if needed.

How is your sump working

It worked great until the side of the tank cracked due to the shelf not being level.
I was planing to rebuild it but picked up an acrylic one.

How is your tank and sump @ +bobyboy

Ended up setting up a Innovative Marine SR80 after this and have been building a large multi-tank 300g total system over the past 6 months. Should be posting a build thread in the next few weeks.
