Hot is on the left and cold is on the right per the Uniform Plumbing Code.

The garden hose Wye is straight hose threads and uses a rubber gasket or O ring as a seal just like a garden hose. The hose thread adapter the RO or RO/DI comes with should also have a gasket or O ring in it as the seal. Make sure to get the wye adapter that has individual ball valves so you can shut the RO off when you are not using it. You should not need to do anything on the washer side and it will remain on all the time.

It has the ball thingy you were talking about and an o-ring at the top.

Also, do I need to tighten the connections with a wrench? Do I need Teflon tape??

Yep that is the one. No, hand tight since the hose gaskets or O rings make the seal and overtightening will cut them. Just like you do a garden hose. No tape or sealants.

It worked! No leaks. Thank you! Tap water was 154tds before. First gallon of ro is 20tds. How low should it get before I add the di part?

you can add the di part right away

You first flush each filter, then add the RO membrane and make 3-5 gallons of RO to waste until the RO TDS is 96-98% lower than the tap TDS. You have quite a ways to go yet before adding the DI. You should see a RO TDS of around 3-5 before installing the DI if you want the resin to last long.
