Just bought home a RedSea reefer 250 g2 and am in the process of getting the plumbing put together now.

We plan on installing both a reef mat and a skimmer on this system, and part of me thinks it would be easier to connect it all now in the very beginning before getting water in the system. Should I plumb it for the reefmat while it's dry, or wait until everything is cycled for a while?

As above. Plumb it dry when it's much easier. You don't need to use the equipment but having it in place will prove the installation and allow you to test.
