I have a one drain one return system and my one drain is a Durso standpipe, should I use a gate valve or no valve becuase it’s just one.

I would bring the return over the back glass and use both holes for drains
Remember tough, Durso's work best with low flow.
Then you would put a valve on the full siphon, slowing flow until a trickle or small stream flows down the other(emergency) drain. It will be silent. It's not like you have to match one with the other.
Using both as a drain, then you can use a valve, and that will help make the drain much quieter.

Meaning the pump will lose some power pushing water up hill.
So durso will probably handle the pump power.
I would skip the durso setup and go for a herbie drain setup.

There are many different types of return nozzles that can be used.
