Welcome to R2R Great bunch of people with a ton of knowledge.
Since you have new sand I would also get some Microbactor7 & add that as well as the Prime. I've used both when ever I make a big water change. If the sand was the Live sand that will help a little with getting the bacteria going but the Microbacter 7 will also help.
