I have a Vecton 25w UV and I want to plumb it off my return pump. Should I run a 1/2 inch to the UV and 3/4 back to the sump? Also would it be ok to put the UV drain in the refugium section to create a little flow? Any help would be appreciated.

What would work for you is a manifold. Basically hard plumb a few valves off the main return to run other equipment. For a UV I would suggest a gate valve for more precise flow instead of a ball valve.

Yes you can put the return of it in your sump, but the big question is can your return pump stand to lose 300gph and keep the rest of the tank flowing nicely.

I am building a new aqueon 125 gallon reef with a fiji cube 30 sump. I will be adding a jabeo mdp 10000 return pump so it would be plenty. I will have two 3/4 returns and plan on teeing off one of them for the UV and teeing off the other for a reactor.

Just my 2cents - i do not plumb anything on return.

Yes plumbing looks easy and some pros like not using extra pump, Hovewer for me lots of downsides:
- my in sump refugium creatures who made into return would be going through UV and back to sump
- if I feed automated frozen while on vacation using return pump if will not be possible obviously
- i will have to fine tune manifold and any little clogs might change the amount of water going into display, meaning yes I will hear it because of emergency overflow and be safe, but I do not like to be in this situation, because reasons for this are plenty and checking which one will be difficult

Instead I would run one extra pipe going into Carbon and UV after for example or another GFO or Algae reactor with return back into sump.

Sure also cons to this method but for some reason it seems more logical.

Just my 2cents.

I run a separate pump for UV. I use a DC pump to control flow. I run the pump in the chamber before refugium so pods have a chance to make it to DT. Also have 2 returns in case a pump goes down

Mis spell - separate PUMP not pipe. And this pump would feed Carbo, GFO, UV and etc.
